Romeo and Juliet Essay Romeo and Juliet is a story about two star crossed lovers. Who fall deeply in love with each outher the moment they meet. At the beginning of the play Juliet is a child like innocent little girl. She does whatever her mother and father tell her to do, I can tell this by when her mother calls her she runs to her mother, and her first words to her mother are madam what is your will? Here she expresses that she is ready to do as she is told. She knows nothing about the world. This is because she has been educated at home. If Juliet does need to go out of the house it is always with the accompany of the nurse or her mother. Juliet is very obedient to her father. The first rapid change we see within Juliet, the innocent little girl. This happens the minute she meets Romeo. She turns really devious. This all happens after Juliet has kissed Romeo, he is about to set off home. Juliet wants to know what his name is, so she asks the nurse, the names of outher young men before actually asking Romeos name. Whats he that now is going out of door? Marry, that I think be young petruchio Whats he that follows there, that would not dance? here she is referring to Romeo. When the nurse does not know his name Juliet sends the nurse to go and ask him. When the nurse returns she tells Juliet his name is Romeo, and a Montague, the only son of your great enemy. This is the first step Juliet takes where she starts to develop her changes from an innocent girl to a young lady who has to make a decision for herself. This is where Juliet has to make a decision whether or not to for get Romeo or to still think of him knowing that he is one of her great enemies. This is also one of the decisions she has to make on her own, without her mother or anyone knowing. Even though she is crafty Juliet is also rather levelheaded. She shows this when Romeo is deeply in love with Juliet and all he talks about in love non-stop. With loves light wings did I oerperch these, walls, for story limits cannot hold love out: and what love can do, that dares love attempt: therefore the kinsmen are no stop to me. At this moment Romeo is just to deeply in love, and does not really think of what he is saying or doing. Whereas Juliet is more down to earth, and reminds Romeo if what danger he could be in. If they do see thee, they will murder thee. It is still the same night she met Romeo, Juliet turns from a child like daughter into a young lady who just does what she feels like doing. This happens when Juliet purposes to Romeo and asks him to send her his answer the next day. At this point Juliet does not really think of what she is doing, Juliet takes things into her own hands and does not bother thinking of the consequences. Juliet does not even ask anyone else before she asks Romeo to marry him, but instead takes matters into her own hands. One other thing that changes about Juliet is that she does not wait for Romeo to bring up the marriage, but brings it up her self. Which is unusual for the girl to be the one that suggests marriage. After taking matters into her own hands Juliet also becomes very demanding, and impatient, something that you would not expect an nai ve girl to be come like in such short time. The clock struck none when I did send the nurse: In half an hour she promised to return therefore here Juliet remembers how long the nurse had told she would take and Juliet had remembered, this also shows that Juliet has kept track of time and has been counting each minute the nurse has been. When the nurse does finally return with the answer Juliet does not lat her sit down knowing she is out of breath, she just wants to her the reply back. But the nurse who just wants to play around with Juliet gives her a hard time, by not telling her the answer. When Juliet says sweet, sweet, sweet Nurse, tell me, what says my love? Repeating sweet three times Juliet sweet talks her way back to the nurse, which makes the nurse to give in. Juliet becomes very romantic and, loving. This is another one of Juliets changes after she meets Romeo. But to be frank and give it the again: And yet I wish but for the thing I have. My bounty is as boundless as the sea, my love as deep: the more I give to thee, the more I have, for both are infinite. Juliet here is saying all this to Romeo and expressing her love to him, so much that she says my love as deep the more I give to thee the more love she gives to Romeo the more I have for both are infinite the more never ending love she holds inside her. Wilt thou be gone? It is not near day. It was the nightingale, and not the lark this is when Romeo and Juliet have spent the night in Juliets bedroom, mourning has broke but Juliet denies that it is mourning when she says it was the nightingale the bird that comes out at night, and that it was not the lark the bird that comes in the mourning. So here Juliet denies that it is mourning because being with Romeo makes time fly past. Therefore this is where Juliet is not the sensible one, and this is the point where Juliet is in an adoring mood. Juliet also shows integrity by saying no to her marriage with Paris. This is where she takes things the hard way. What she could have done she could have married Paris and forget all about Romeo as he was banished. But she loved Romeo and took the hard way. There is another frantic change in Juliet. Being with Romeo has also made her very brave. The first brave aspect we see in Juliet is when she marries her fathers enemys son, Romeo. Knowing what can happen to Romeo and her she still takes the big step in marring Romeo. She is also exceedingly brave when she tells her father that she will not marry Paris. Not proud you have, but thankful that you have. Proud can I never be of what I hate, but thankful even for hate that is meant love. This is the first time ever she has disobeyed her father. Making her point loud and clear in saying no to marring Paris. She is also very brave when she takes the potion. At first she cant wait to get her hands on the potion, give me give me. Knowing the potion can go wrong and if she does wake before Romeo has got to her. When I am laid into the tomb, I wake before the time that Romeo comes to re deem me. She still takes the potion for her loves sake. Again she is extremely brave when she kills herself when she sees Romeo dead on the floor Juliet sees the dagger as some sort of cover that will protect her from everything that is happing and take her to her Romeo O happy dagger! This is thy sheath, there rust, and let me die. On the whole I think Juliet does what she does because of the difficulties this feud has caused for her relationship with Romeo. I also think that it is good that Juliet changed throughout the story because if she did stay as an innocent girl she would have never gone as far as marring Romeo. Throughout the whole play every change that she made helped her in many ways. If Juliet did not turn crafty or disloyal to her father she would have never ended up dieing with Romeo. GFP Practical Report

GFP Practical Report GFP is very useful as a reporter protein. After its discovery in 1962 its practical applications were put into use 30 years later by adding the coding DNA of GFP before the stop codon of other proteins. This allows for an easily detectable marker of the proteins presence without needing additional cofactors or causing any harm to the organism. The spectral characteristics of GFP can be changed by making mutations to the protein. In this investigation a Y66W mutation was made to wildtype GFP in order to produce a shorter excitation and emission wavelength. The mutation was made using QuikChange site directed mutagenesis. The protein was then cloned into BL21(DE3) pLysS for expression. The cells were then lysed and applied to a Ni-NTA column. This fractionated the lysate in order to analyse these fractions using SDS-PAGE, fluorescence and Bradford assays. It was found that the Y66W mutation was successfully added but due to another mutation in the stop codon additional amino acids were added to the C terminus of the protein. It was also found that purification was partially successful as GFP was eluted in the correct fraction. This is supported by the Bradford and fluorescence assays. The green fluorescent protein (GFP) is a 238 amino acid protein with a molecular mass of 26,870 Da. It was first isolated from the jellyfish species Aequorea Victoria by Osamu Shimomura in 1962 (1). GFP is expressed in small photoorgans that are situated in the umbrella of the jellyfish. Douglas Prasher first realised the potential of GFP as a reporter protein (2). As proteins are smaller than the resolving power of electron microscopes, Prasher thought the GFP gene could be added into the gene for haemoglobin before the stop codon. This would allow the protein of interest to maintain all of its functions but would have the GFP protein at its C terminal end. This means that detection of GFP fluorescence would also indicate the presence of haemoglobin. Furthermore, GFP does not require additional cofactors or substrates to fluoresce. This means that it works extremely well as a non-invasive method of detection of protein expression. GFP is also non-toxic so it is able to be used in vi vo without causing damage or harm to the organism. Crystallisation studies (3) have shown that GFP has a barrel structure with the chromophore buried in the centre. This chromophore is comprised of 3 amino acids (Ser 65-Tyr 66-Gly 67) that undergo a series of spontaneous cyclisation reactions to create the active chromophore. Wild type GFP has a major excitation peak at 395 nm and a minor one at 475 nm with an emission peak at 509 nm. In vivo GFP is coupled to the protein aequorin which induces a blue glow when it interacts with Ca2+ ions and breaks down luciferin. This light is able to excite GFP and cause fluorescence. In vitro this is not the case, however GFP fluorescence can be easily induced by irradiating GFP with UV light. As with all proteins, GFP can be mutated. By mutating key residues, such as residues in the chromophore, it is possible to change the characteristics of GFPs fluorescence. The first of many mutations was the S65T mutation (4). This mutation improved the characteristics of the protein including increased photostability, fluorescence and a shift of the major excitation peak. This investigation is based on the engineering of GFP to create a mutant of GFP with a shorter excitation and emission wavelength by inducing the Y66W mutation. The aims of this investigation were as follows. To carry out site directed mutagenesis of GFPuv to clone into pET28c and transform the products into XL-1 super competent cells. Extraction of the plasmid after incubation overnight to check the purity and concentration of DNA. Preparation and transformation of BL21(DE3) cells. Lyse these cells and fractionate the lysate to purify his tagged GFP using a Ni-NTA column. Finally, detection of purified GFP by SDS- PAGE, Bradford assay and fluorescence. The workflow of the investigation can be found in figure 1 in appendix 1. A more detailed protocol can be found in the BIOC2302 semester 2 practical manual on pages 6-15 with rationale for all experiments. In site directed mutagenesis I 31 ÃŽ ¼l of water was added to the PCR reaction to give a total reaction volume of 50 ÃŽ ¼l. In site directed mutagenesis II a supplied culture of cells was used in the experiment rather than cells from the transformation colonies in site directed mutagenesis I. His tagged GFP was used instead of the mutant in the protein purification experiment in order for easier administration as the process is the same. Site directed mutagenesis I Before the wet lab work began it was first necessary to design primers for QuikChange to induce the Y66W mutation into the wild type GFP. These can be seen as figure 2 in appendix 2. These were created using the QuikChange primer design tool on the Agilent website. The site directed mutagenesis was carried out using the primers supplied to induce the correct mutation. The products of this were cloned into the pET28c plasmid and the XL-1 super competent cells. The cells were plated as per the BIOC2302 practical manual and left to incubate overnight. Site directed mutagenesis II Upon checking the plates in the next session it was found that no transformed colonies had grown so a new culture was supplied. The undigested plasmid control grew approximately 50 colonies The culture of BL21(DE3)pLysS cells was set up and the OD600 were recorded. They can be seen in table 1 in appendix 3. Within 50 minutes the culture had reached an OD600 of 0.483 meaning the cells were at the correct density for lysis. The cells were prepared as per the BIOC2302 practical manual and the recombinant plasmid was extracted. The concentration measured was 121.7 ng/ÃŽ ¼l and the A260/A280 was 1.86 using nanodrop. Therefore, the ethanol precipitation was not carried out. To prepare for sequencing 4.11 ÃŽ ¼l of this solution was diluted, with 5.98 ÃŽ ¼l EB buffer, to the correct concentration. This was then sent to be sequenced, the results of which can be seen in appendix 4. The primer has been highlighted in green and is surrounded by a box with the mutated codon in red. A deletion also occurred in the stop codon of the mutant as highlighted by the second box with deleted bases highlighted in blue. Protein purification The plates were inspected in the next session. It was found that the 200 ÃŽ ¼l transformation plate grew 3 colonies and the 50 ÃŽ ¼l transformation plate grew none. Transformation efficiency can be calculated for the 200 ÃŽ ¼l plate as 37 transformants/ÃŽ ¼g of DNA. The cells were weighed and found to be 0.539 g so 2 ml BugbusterTM used. After lysis and fractionation the SDS-PAGE samples of each fraction were prepared and loaded onto the gel. The Bradford assay was carried out while the gel ran.   The BSA standards were calculated and the contents of each standard well can be seen in table 2 in appendix 3. The fractions were then diluted into their wells and the contents can be seen in table 3 in appendix 3. The plate was filled according to the map in figure 2 in appendix 5. The plate was ran and the absorbances for the BSA standards were taken from the plate readout and inputted into table 4 in appendix 5. From here a calibration graph was set up using GraphPad Prism and can be seen as graph 1 in appendix 6. This graph shows that the data points for the standards do not fall near the line of best fit. The absorbance results from the plate readout for all of the fractions were imputed into table 5 in appendix 7. The equation of the line from graph 1 was then used to calculate the concentration of protein in each of the fractions. All of these values were also inputted into table 5. With the Bradford assay complete the SDS-PAGE gel was disassembled, stained and a picture was taken. A map of the gel can be seen as figure 3 in appendix 7 and the picture of the gel can be seen as figure 4 in appendix 8. By looking at the picture it can be seen that in lanes 2, 3, 4 and 9 there are dark bands spanning the entire lane. In 5, 6 and 8 there is faint banding across the well. In well 7 there is a distinct small band in between the 25 kDa and 37 kDa molecular markers. Lane 8 shows no bands at all. Finally the fluorescence assay was carried out as per the map of the microtiter plate in figure 5 in appendix 7. The results from the plate readout were inputted into table 5. From here a graph comparing the log of protein concentration compared to fluorescence of each fraction was plotted and can be found as graph 2 in appendix 6. This shows elution 1 with the highest fluorescence and the unbound x10 had the lowest. However, when comparing protein concentration the unbound fraction had the highest and wash 2 had none. Percentage fluorescence was also calculated and inputted into table 6 in appendix 9. The first aim of this experiment was to transform the site directed mutagenesis products into XL-1 super competent cells. The correct primers were used in order to induce the Y66W mutation into the parental DNA. However, no colonies that were meant to take up the mutated plasmid grew but the undigested control grew around 50 colonies. This means the cells did not take up the plasmid because otherwise they would have grown on the plate. This could be due to a mistake made in making the PCR reaction mixture or the DNA may have become damaged at some point in the experiment. Additionally, the suppliers of the XL-1 super competent cells advice to avoid large changes in temperature. This was unavoidable in this experiment and may have contributed to the cells not taking up the plasmid. In the future more care should be taken while plating and preparing the cells. Also preparation of any reaction mixtures should be checked very closely in order to ensure the correct reactants are added in the correct amounts. In site directed mutagenesis II the cell culture was lysed when the OD600 was 0.483. That is because E.coli cells are most likely to be made competent when they enter early log phase. This corresponds with an OD600 of 0.4-0.5. The DNA concentration extracted in this experiment was found to be 121.7 ng/ÃŽ ¼l and an A260/A280 of 1.86. This means that the DNA is good quality as the desirable range for A260/A280 is 1.7-2.0 and the concentration was much higher that what was required. However, in future experiments to test for reliability multiple results should be taken. Furthermore, the data could have been confirmed by using the spectrophotometric method alongside using nanodrop. The sequencing results in appendix 4 confirmed the successful incorporation of the Y66W mutation into GFP, creating the CFP mutant. However, the second mutation at the stop codon deleted 2 bases including the first base of the stop codon. This means that when the protein is expressed the ribosome will not stop and instead will continue to add amino acids onto the C terminus of the mutant until it reaches a new stop codon. There 144 bases between the original stop codon and the next in frame stop codon meaning 48 additional amino acids will be added to the C terminus. This codon can be seen highlighted in purple below the original stop codon. These additional amino acids could affect the folding or could increase the likelihood of aggregation of the mutant protein. In the protein purification experiment the 200 ÃŽ ¼l transformation plate grew 3 colonies and the 50 ÃŽ ¼l transformation plate grew none. The transformation efficiency on the 200 ÃŽ ¼l plate was 37 transformants/ÃŽ ¼g of DNA. The reason why this is so low could be due to a number of factors such as the plating technique or the cells may not have been left to chill on ice for the optimum amount of time. However, the negative control did not grow any colonies, confirming that all of the bacteria on the transformation plate were transformed. Again, more steps should be taken if this was to be carried out again to ensure that proper plating and prepping protocol is followed. The Bradford assay shows that in wash 2 there was no protein in the well. This means that any protein found in elutions 1 and 2 should all be His tagged GFP that bound the Ni-NTA column. This can be confirmed by the fluorescence results as the elution 1 fraction contained the majority of the total fluorescence with 44.13% of the total. However, all other fractions also produce some fluorescence. This could be due to GFP contamination in the other fractions. This could have occurred due to the resin being saturated, preventing further binding to the column. It could also be due to aggregation of the protein obscuring the His tag and preventing binding. Furthermore, the plots on the calibration graph do not fall on the line of best fit. This means that the equation of the line is not accurate and protein concentrations calculated using it are also inaccurate. Therefore, there could be more protein in each fraction than was calculated. This could account for the fluorescence in the wash and unbound fractions. The Bradford assay is quite limiting. This is due to the fact the assay only measures protein concentration rather than GFP concentration. This means that it is unsure whether the protein concentration measured in elution 1 and 2 is all GFP or it is contaminant protein. The same can be said for the other fractions, its unsure whether the protein concentration measured has been contaminated by GFP. In the future this assay should be carried out again to try and reduce contamination. The calibration graph should also be repeated until all of the data points fall on the line of best fit. Otherwise none of the calculated protein concentrations are accurate. Finally, the SDS-PAGE results shows banding in wash 1, 2 and elution 1 and 2. This suggests that there is contaminating protein in all of these fractions. Elution 1 shows a clear band at approximately the 26-27 kDa mark as it is present just above the 25 kDa marker and is well below the 37 kDa marker. This suggest the band in elution 1 is GFP as it is the appropriate size and is in the expected fraction. Another source of error could be due to the amount of pressure applied to the pipette. This will vary from person to person and will affect the volume of the solution being pipetted. As such small volumes were being used and there was a lot of solutions to be pipetted it is very possible a mistake was made. This mistake would have a big effect on the concentration and therefore could have a big effect on the absorbance values. These errors can be avoided in future by using the appropriate pipette for the volumes being used. Further reduction in errors can come from correct technique and by doing replicates and averaging values. There could be some error in the microtiter itself. There may have been markings or scratches on the plate that werent seen at the time. This could affect how the light passed through the reader and therefore affect the absorbance values. In conclusion, the aims of this investigation were to induce the Y66W mutation into wild type GFP using QuikChange site directed mutagenesis. Furthermore, the protein was to be expressed in competent BL21(DE3) pLysS cells. Finally wildtype GFP was to be purified using a Ni-NTA column and the fractions analysed with SDS-PAGE, fluorescence and Bradford assays. The investigation successfully introduced the Y66W mutation into wildtype GFP. However, the stop codon was also mutated adding an extra 48 amino acids on the C terminal of the protein.   A band indicating the presence of GFP was found at the 26.9 kDa mark in elution 1, indicating it was bound to the column and was eluted. Creons Role in Antigone :: Creon as tragic figure

Creon in the play of Antigone by Sophocles plays a major role within the play. Antigone also plays an important role, as these two character’s conflicting views led to utter disaster, which highlights Creon as a tragic figure. Within the play Creon attempted to establish decisions for the common good; however, his decisions resulted in tragedy. Creon highlighted as the tragic figure, initially created decisions he thought was for the welfare and well-being of Thebes; however, Antigone, who rebelled against Creon’s decisions, caused Creon’s rage to cloud his rational way of thinking. When Creon decided to forbid the burial of Polynices, as he believed Polynices was a traitor to Thebes, Antigone was outraged. This decision, viewed through the eyes of Creon, was just and fair; on the other hand, Antigone viewed his decision as cruel and selfish, which resulted into a major conflict between these two characters. When Antigone disobeyed his proclamation, Creon became infuriated towards this rebellious act. Those small events within the play expressed Creon as a ruler doing what he believes is right; on the other hand, Antigone’s rebellion expressed otherwise. Antigone was soon shown to cause an evil spark within Creon. This â€Å"spark† caused Creon to intensify his punishments towards Antigone, which in turn caused a chain reaction of events and eventually led to his tragic decline. Being portrayed as a menacing villain throughout the play, Creon began as an honorable man; however, overwhelming events and confrontations caused Creon to evolve in to this monstrous figure. This play, Antigone, was titled after one of the play’s main characters; however, the title could have been â€Å"Creon†, due to Creon’s influence throughout the play. Creon was a major influence to the play’s plot as it involves Creon within a majority of the scenes, from beginning to end. Throughout the play Creon’s tragic countdown is expressed, beginning with him as king of Thebes to the death of his beloved family. Antigone is merely portrayed as a rebellious nephew, as she consistently defies Creon. Antigone is expressed within the play consistently; however, the play portrays decisions proclaimed by Creon, which displays Creon’s tragic flow. Being titled â€Å"Antigone†, does not accurately express the play’s content as Creon’s tragic fate is actually being described and represented.

Human Rights Violations Against Children Essay -- Universal Declaratio

As human beings, children are entitled to all the rights guaranteed by the Universal Declaration of Human Rights (UDHR) and the various treaties that have developed from it. But children also need special protection and care. They must be able to depend on the adult world to take care of them, to defend their rights and to help them to develop and realize their potential. Yet, violence against children is endemic: each day, terrible abuses and acts of violence against children are committed worldwide. They suffer as many of the human rights abuses as the adults, but may also be targeted simply because they are dependent and vulnerable. The Fifth Article of the Universal Declaration of Human Rights states that "no one shall be subjected to torture or to cruel, inhuman or degrading treatment or punishment". Nevertheless, children are being tortured and mistreated by state officials; they are detained, lawfully or arbitrarily, often in appalling conditions; in some countries they are subjected to the death penalty. Countless thousands are killed or maimed in armed conflicts; many more have fled their homes to become refugees. Children forced by poverty or abuse to live on the streets are sometimes detained, attacked and even killed in the name of social cleansing. Many millions of children work at exploitative or hazardous jobs, or are the victims of child trafficking and forced prostitution. Because children are "easy targets", they are sometimes threatened, beaten or raped in order to punish family members who are not so accessible. Amnesty International has been one of the organizations that has denounced this terrible situation in a new report published prior to the Human Rights Day. The report of Amnesty International sho... ...taining 46 children. The children fled outside where they were made to lie face down on concrete, some only in their underwear, for hours. Several were allegedly sprayed in the face with mace while on the ground. The memorandum in support of the injunction noted that "penal officers at Jena have rubbed inmates' faces into cement floors, taken away clothing, slammed youths against doors, walls, and floors, and forced naked juveniles to squat with their buttocks in the air while searches are performed ... evidence exists showing officers actually have encouraged peer violence." The situations mentioned above also apply to other institutions such as orphanages and refugee centers that in addition can be vulnerable to a great deal of exploitation by being used like subjects in drug experimentation and undergo cruelty, negligence, confinements and corporal punishment.

Philosophers of the Age of Enlightenment Essay -- European History

During the eighteenth century, ideas of reform started in France and spread through Europe. This period is referred to as The Age of Enlightenment. The Enlightenment carried the idea that economic change and political reform were possible. People started to think that they could use their own intellect to challenge the intellectual authority of tradition and the Christian past. The people who wrote for change and reform were called the philosophes (French for philosophers). They wrote hoping to bring reform to religion, political thought, society, government, and the economy. Thanks to the print culture, the philosophes’ ideas were spread throughout Europe. People started to have educated conversations about these new ideas in places like coffeehouses and drinking spots. The ideas of philosophes during the Enlightenment challenged whole societies. Some of philosophes include Denis Diderot, Jean Le Rond d’Alembert, Immanuel Kant, Adam Smith, Montesquieu, Jean-Jacques Rou sseau, and Voltaire. In the mid eighteenth century, the Encyclopedia was published. It was written under the leadership of Denis Diderot and Jean Le Rond d’Alembert. Over one hundred French thinkers contributed to the Encyclopedia, including all of the major French philosophes. Diderot wrote on about every subject including philosophy, science, music, and art. D’Alembert divided and described the different sciences, and he analyzed the role of reason. The Encyclopedia was meant to secularize learning and to refute intellectual assumptions from the Middle Ages. It contained articles on religion, government, and philosophy. It attempted to see the natural world through science and technology, and human passions through an understanding of how individuals and societies work. The Encyclopedia helped spread the thoughts of the Enlightenment over the continent. Immanuel Kant was an important German philosopher who coined the term the â€Å"Enlightenment.† He said, â€Å"Enlightenment is man’s emergence from his self-imposed nonage. He was one of the first philosophers in the Enlightenment, and some of his ideas lead to the French Revolution. Denis Diderot, Jean Le Rond d’Alembert, Immanuel Kant, Adam Smith, Montesquieu, Jean-Jacques Rousseau, and Voltaire all helped to write about possible reform for society. MRP (Materials Requirements Planning)

From reading about material requirements planning (MRP), I deciphered that this is a manufacturing planning and inventory control system that helps manage manufacturing procedures. MRP systems ideally have three main purposes.The first is to make sure that all materials are obtainable for manufacturing and the products are accessible to deliver to the buyers. The second purpose is to uphold the low inventory in store, and the third is to design manufacturing actions, delivery agendas and purchasing actions.The simple purpose of MRP system is to regulate the inventory. The use of MRP assists companies in keeping their inventory as low as possible while helping plan manufacturing, purchasing and delivering functions.Businesses need to regulate the kinds and amounts of supplies that they buy, strategize which products are to be made and how much of it and confirm that they are capable of meeting existing and upcoming consumer requests, while sustaining the lowest price for the company. Making a bad decision in any of these areas will make the company lose money.Not planning accordingly within this criterion could cause the company to lose a lot of capital. MRP can help you know what resources are needed and how many, as well as when you will need these materials.MRP can be used for materials that are bought from external sellers as well as those made in-house. You need to keep in mind what materials you are making, how much of it do you need, how long they can be stored for. For the planning portion, you need to know what limitations you may have for producing the materials as well as how to make them.For the outputs and reports part of MRP, you have output 1 which is the â€Å"Recommended Production Schedule†. What this does is that it gives you a thorough schedule of the dates that you need to complete the task along with how many to fulfill the call from the Master Production Schedule.Output 2 is the â€Å"Recommended Purchasing Schedule†. This ou tput gives you the dates you will get the materials bought and the dates that the purchase orders should transpire to equal the production schedules. Like with any other system, there are problems that evolve from using theMRP system. The main issue with MRP systems is the integrity of the data. If for some reason there are any mistakes in the inventory data, the bill of materials data, or production schedule, then this would make the output data wrong.To rectify these issues however, some businesses use pull systems and bar code scanning which makes it more accurate. A big concern with the MRP systems is the fact that they are mandated to state the length of time that a manufacturer will need to make a product.The system assumes that this time in manufacturing won’t change every time the item is produced, with no concern as to how many are made, or other things that may be made at the same time. Regardless of some of the issues that the MRP system may have, it does help comp anies reduce cost and inventory on hand.It helps companies produce what they need, when they needed to meet any demands that any consumers may have as well as deadlines etc. This system works well with many different types of companies and it seems to be fairly efficient.

Psychological Perspectives for Health and Social Care Essay

Psychologists uses a range of perspectives and approaches when studying how individuals think, feel and behave. Some researchers may focus on one specific perspective, whilst other researchers study a more diverse approach that may incorporate multiple points of views. Each perspective aims to offer explanations for different aspects of human behaviour. The behaviourist approach’s influence to health care The behaviourist approach is based on the concept of explaining behaviour through observation and the belief in which our environment is what causes us to behave differently. The behavioural learning model learning is the result of conditioning. The foundation of conditioning is that a reward following a desirable response performs as a reinforcer and increases the possibility that the desirable response will be repeated. Reinforcement is said to be the core of the behaviourist approach. Furthermore, once a desired behaviour established, irregular reinforcement maintains the behaviour. The behaviourist theory approaches are frequently used in weight loss, smoking cessation, assertiveness training and anxiety-reduction programs. The significance of frequently and consistently rewarding desired behaviour immediately and not rewarding undesirable behaviour is crucial to the success of a behaviourist approach to learning. The principles of classical conditioning have been applied in many therapies. As its name suggests, behavioural therapy is focused on human behaviour and looks to eliminate unwanted or abnormal behaviour. Typically this type of therapy is used for those with behavioural problems or mental health conditions that involve unwanted behaviour. Examples of this include: addictions, anxiety, and methodical desensitization for phobias, aversion therapy and obsessive-compulsive disorder (OCD). Practitioners of behavioural therapy believe that behaviour is learned and can therefore be un-learned through therapy. As well as the behaviour  itself, behavioural therapists will look at thoughts and feelings that lead to the behaviour or occur as a result of the behaviour to comprehend the issue at a greater level. Aversion therapy is a form of treatment that utilizes behavioural principles to eliminate unwanted behaviour – as it follows, if all behaviour is learned it can be unlearned. In this therapeutic method, the unwanted stimulus is repeatedly paired with discomfort. The objective of the conditioning process is to command that the individual associates the stimulus with unpleasant or uncomfortable sensations. There are many reasons why behaviour could perhaps be unlearned; this could be due to behaviour in which is destructive or undesirable. These undesirable behaviours come about as individuals associate them with pleasure; the brain learns that, such as, drinking may allow one to feel relaxed a lowers stress levels. This is somewhat fine, however if one becomes reliant on the substance and it begins to take a dominant part in one’s life then this has become an undesirable behaviour. It is one’s choice to unlearn that alcohol equals pleasure. Aversion therapy goes about eliminating this behaviour by attempting to break the association between alcohol and pleasure. The therapy, in the case of alcoholism, involves the patient drinking while together having a negative stimulus directed. The negative stimulus could be an emetic drug (one that causes the patient to vomit when drinking alcohol) such as an emetic drug, one that encouraging vomiting when alcohol is consumed like disulfiram (a synthetic compound used in the treatment of alcoholics to make drinking alcohol produce unpleasant after-effects), or an electric shock administered whenever the patient drinks. In short, then he patient is punished for drinking and, for the same reason a parent punishes a child, a successful outcome is to reduce or completely eliminate their undesirable behaviour. The cognitive approach’s influence to health care Cognitive therapy for depression has its roots in the cognitive theory of depression (Beck, 1967). It is an active, structured, problem-focused, and  time-limited approach to treatment which is based on the premise that depression is maintained by negatively biased information processing and dysfunctional beliefs. Treatment is designed to help patients learn to think more adaptively and thereby experience improvements in affect, motivation, and behaviour. The effectiveness of cognitive therapy for depression has been demonstrated in over 30 clinical trials (Dobson, 1989). The general approach in cognitive therapy for depression involves guiding patients through a number of structured learning experiences. Patients are taught to monitor and write down their negative thoughts and mental images to recognize the association between their thoughts, feelings, physiology, and behaviour. They learn to evaluate the validity and utility of these cognitions, test them out empirically, and change dysfunctional cognitions to reflect a more adaptive viewpoint. As therapy progresses, patients learn to identify, evaluate, and modify underlying assumptions and dysfunctional beliefs that may have predisposed them to depressive reactions. The therapist also teaches (or reactivates) adaptive coping skills such as breaking down large problems into smaller, more manageable steps, and decision-making by cost-benefit analysis. Activity scheduling, self-monitoring of mastery and pleasure, and graded task assignments are commonly used early in therapy to help patients overcome inertia and expose themselves to potentially rewarding experiences. Patients typically require approximately eight sessions to gain a reasonable level of mastery with the model and the skills involved. A significant reduction in symptoms often occurs during this initial stage of therapy. The remaining sessions are used to evaluate and modify dysfunctional beliefs that impair functioning and make the patient vulnerable to future depressive episodes, build relapse prevention skills, and discuss termination issues. According to my research, many patients show a remission of symptoms in 8-12 sessions. A full course of treatment is considered to be 14-16 sessions although severe cases can take longer. Maintenance of treatment gains is enhanced by occasional booster sessions during the first year after one’s termination. The humanistic approach’s influence to health care Humanist learning theorists view learning as a function of the whole person and believe that learning cannot take place unless both the cognitive and affective domains are involved. The individual’s capacity for self-determination is a vital segment of the humanist theory. For example, the humanist theory is used to help post myocardial infarction (a syndrome that involves the inflammation of the sac surrounding the heart); patients regain a sense of personal control over their health care management. The focus of the humanistic perspective is on the self of one individual – which translates into you, and your perception of your individual experiences. This approach argues that one is free to choose his own behaviour, rather than responding to environmental stimuli and reinforcers. Issues dealing with one’s self-esteem, self-fulfilment, and requirements are seen as dominant. The key focus is to assist one’s personal development. Two major theorists associated with this view are Carl Rogers and Abraham Maslow. All patients grow with success and do better when achievements are recognized and reinforced. Respecting the whole person in a supportive environment can encourage learning. Learning is also fostered through structuring information appropriately and presenting it in meaningful segments with appropriate feedback. There are a vast variety of conditions that should be encountered before an individual can develop on becoming self-actualized. According to the ‘needs hierarchy’ described by Abraham Maslow, individuals must first secure their basic â€Å"organismic† needs (including adequate food, clothing and shelter necessary to keep them alive). Having achieved the essentials, they next build up and work to achieve: a feeling of adequate safety, a sense of belonging (to one or more social groups and relationship), and a sense of self-respect and social respect. Self-actualization, the drive for one to do all that he desires to do with his life, is something that only occurs as a influence of behaviour after all the earlier needs are adequately satisfied and a state of contented happiness is achieved. For instance, the media create unrealistic, and for most individuals  unattainable ‘ideal’ image, especially for women and adolescent girls. The majority of models exposed publicly are greatly below the ‘normal’ weight for their age and height. In the humanistic vision, human dysfunctions are caused by a faulty or interrupted development process; essentially human issues regarding to immaturity, or commonly of the social/emotional variety. The aim of humanistic therapy is to promote social or emotional maturity and growth. Through assisting service user’s to resume their disrupted developmental processes in healthy directions, patients are helped by professionals in order to grow up and out the of the immature mental and emotional states that contribute to the pain one may feel or cause pain upon others. The psychodynamic approach to health care Anxiety is a feeling of worry, extreme nervousness, or unease about something with an uncertain outcome. The condition gives of an uncomfortable feeling of fear or an approaching disaster and could perhaps negatively reflects the thoughts and bodily reactions an individual may encounter when presented with a situation that is unable to be managed. When an individual experiences the feelings of anxiety, their thoughts may often actively assess the different situations without intentionally doing so; the individual may too develop predictions of how they will cope founded on past experiences. Despite the fact that some anxiety is a normal response to difficult and stressful circumstances, whereas the anxiety level is abnormally high an individual may lack the awareness of how to effectively control the issue. Anxiety can take many forms, and several of these may consist of: An intense physical response due to the arousal of the nervous system leading to the physical symptoms (which may involve the racing of a heartbeat). A cognitive response referring to the thought about the issue and the individuals ability to manage with it. Those which encounter the condition of anxiety may often feel negative about most situations and think unenthusiastic thoughts. A behavioural response which could consist avoidance or unusual behaviour including aggression, restlessness or  irrational behaviour. An emotion response reflecting the high level of distress the individual is confronted with. There is just not one cause of anxiety, however there are a number of factors that could contribute to the development of anxious thoughts, feelings and behaviour. The many factors comprise of: Hereditary – many research has suggested that those with a family history of anxiety are more likely to also develop anxiety. Biochemical reasons – Research suggests that individuals who experience a high level of anxiety may have an imbalance of chemicals in the brain that regulate feelings and physical reactions. Certain life experiences – Particular life experiences can allow individuals more vulnerable to anxiety.
